Head to the Skyfrost Nail waypoint location marked in the map below and you will find the Secret room. To open the sealed gate, you have to use the Princess Box, Priest Box and Scribe Box that you have found from all over Dragonspine.
Upon opening the gate, you can loot some chests inside. This will get you a Crimson Agate and a Stone Tablet as well.
